Only on Labor Day for the Labor Day Bridge Walk, for which the Bridge is closed to all vehicles. The Walk begins at about 6 am, and the last entry is between 10 and 11 am, so that the Bridge is cleared for traffic. But be sure to book your lodging early in the year; everything in the area is pretty much full by June. The DALMAC (Dick Allen Lansing to Mackinaw City) bicycle tour used to cross to St. Ignace the Sunday before Labor Day, but the final year was, I think, 2016. Now the only time you can cross outside of a motorized vehicle is during the Labor Day Bridge Walk - mostly for safely concerns. The wind gusts running through the Straits can get pretty intense, causing traffic to slow down under the posted speed limit of 35 mph. It would be easy for a pedestrian to get blown off. Also, the Bridge is about 5 miles from land anchor to land anchor. That's a long way to walk.

What makes the Bridge Walk different? Lots of other people (in the thousands) and lots of emergency personnel, including State Police from across the state. I suspect the Mackinac Bridge Authority would rather not have the Bridge Walk (likely for liability reasons); but it has become such a Michigan tradition and is such an economic boon for the area (local business viability for the next year literally relies on the Walk participants) that it can't be stopped.
