Affinity Chromatography is currently the most widely accessible and user-friendly technique for protein purification, esp. recombinant proteins. It relies on the "affinity", i.e., binding ability of a protein to a small molecule known as a ligand.
Using this technique, proteins which bind to DNA, RNA, Carbohydrates, lipids, or any other ligand can be easily purified.
I discuss the principle of Affinity Chromatography along with the related derived technique of TAP-tagging, which is useful to isolate protein complexes.
