Amazing Profesor! I was very dissapointed that I could not do this experiment because of the COVID-19. Although I was very exited and the class was amazing!! Hope to see more videos!!

Thank you, thank you, thank you for doing this video. I was so incredibly disappointed that my students are not able to do this wet lab. They love seeing their bacteria glow. Surely students handling say e-coli would give the disinfectants industry such a boost. And so the pharmaceutical companies for anti -biotics ! Imagine what fun it would be if she "just snipped out " an Antibiotic resistance gene " and shoved that in there !. - or a "covid 19" spike protein " gene. Then call it a "vaccine" and tell all the "mice" that it will "protect them" if injected into the shoulder muscles !. No even better MANDATE that they must do that or everything they own is taken away for public health safety. Yes that's sure to be very popular - with pharma shareholders, CEO's etc. who will then all be worshiped and given enormous pay rises for "saving the world from the terrible Thing from "Wuhan", for instance! And all the green ladies with horrible falceto voices can tell any survivors how it was done - assuming that they all "vaxxinated" themselves with the Thing resistant gene.

we performed this experiment and it was very disapointing since we didnt get the result of a glowing bacteria E.coli. we left it overnight and during the morning when we place our petri dish under UV light there where no signs of bacteria culture in LB/AMP and AMP/ARA. what could be the cause of this, since you said that the plasmid that has a GFP genes does not affect the ampicilin.But in pur petridish there were no bacteria?.

Thank you for your question! As it turns out, there are three main types of UV light: UV-A, UV-B, and UV-C. UV-A has the longest wavelength, and therefore, has the least amount of energy. UV-A is the type of UV light produced by what we commonly refer to as "blacklight"--and thus is safe for Ms. McKechnie to handle as she does in the video. UV-B and UV-C type UV lights, which have shorter wavelengths, have much more energy, and are viewed as mutagens (they have so much energy that they can damage or mutate your DNA in harmful ways). When we use those higher energy UV lights, we have UV-blocking covers to protect us and others in the lab from the harmful effects of UV exposure. We sometimes also use face shields, or protective eyewear, depending on the light source and how it is being used.
