For anyone watching, prophylactic use of antibiotics is not acceptable in most labs. Often it only keeps contamination below detectable levels and can lead to antibiotic resistance. Good aseptic practice is the only defense needed to keep contamination free cell cultures, and if contamination does arise simply pull from your seed stock.

Yes, the addition of the complete medium containing serum (FBS) inactivates trypsin with its several antiproteases

Where do the cells actually come from? I have never understood where the lab actually gets them from. They're always just already in media being grown at 37C when I see a demonstration.

there is always one cell. for example for IVF we just fertilize the sperm and the egg to make zygote and then after a day it would be a cell and it would grow up...

as other have pointed out, there are cell banks. you go online, pick your cell line (no small task), buy it, it arrives, your lab stores keeps it at the relevant temperature, you pick it up, you whack it in some appropriate media and bam! sometimes there are additional steps to induce phenotype, differentiation, profileration etc

It requires certification for certain biosafety levels for you to order cells (often from ATCC). They will arrive probably frozen and can be thawed. Some labs make their own cell lines from primary cells or other cell lines.
