IB students that missed an ISC class here are the answers - What are the pigments and why do plants and algae need them? The pigment of the plant or algae shows which colors they will absorb from electromagnetic radiation and the colors they will reflect - Explain the materials needed to conduct the experiment - Piece of coffee filter - 15ml of 95% ethyl-alcohol - A pinch of sand - 2 toothpicks - A mortar and pestle - 50 ml graduated cylinder - 250ml beaker - Test tube and rack - Plastic wrap - Ruler - Pencil - Scissors - 5-10 fresh spinach leaves - Briefly explain the method of the investigation - Cut the leaves into piece with scissors - Grind the leaves with the pestle for a minute - Add a pinch of sand while grinding - Add 10 ml of the ethyl-alcohol - Mix for another minute - Place the mixture in the 250ml beaker - Cover the top with plastic wrap - Leave for 10 minutes - Cut a strip of the coffee filter and make sure it’s longer than the test tubes - Roll an end of the paper around the tooth pick - Place the toothpick with the paper wrapped around in the test tube, with the toothpick resting on top - The paper should not touch the bottom - Take the paper out and measure 1-2 cm of the bottom end of the paper and draw a line across - Take the other toothpick and dip it into the mixture and place the sample onto the line previously drawn - Pour 5 ml of ethyl-alcohol into the test tube - Place the filter paper with toothpick back into the test tube and cover the whole thing with plastic wrap - Leave for 20 minutes - Remove the paper and let it dry GL bros

Fantastic stuff. I really liked the end where you suggested that leaves of different colo(u)rs are worth looking at and also about the possible relationship between levels of chlorophyll and growth rates. To be honest, with a little bit of variety, this simple qualitative experiment could be easily modified into a detailed experiment covering the 90 minutes that I have to do chromatography with some visiting junior high school students (here in Japan). Thanks for the great video and ideas.

My teacher recommends Kale, Lettuce, Spinach, Collard Greens, or Romaine Lettuce. But you should avoid using endives, iceberg lettuce, or leeks.

Why a plastic wrap? Is it so that the solution is not exposed to air? But whats gonna happen if it is? Will it not work then? Im asking this because its really hard to find a plastic wrap like that one where I live.

I would say the ethyl alcohol, which has a boiling point of ~78 °C, would evaporate and leave the exposed test tube. Ethyl alcohol is flammable and poisonous with skin contact or ingestion in its pure forms. So if you plan to do this, do it under caution.
