In this final installment of the Learning Electroporation series, I address some of the key issues plaguing past failures and low electroporation transformation rates. We'll visit the topic of low-copy plasmids, running silica-based minipreps that target low-copy plasmids, estimating dna concentration and finally, the protocol I developed that finally yielded positive transformation rates.
With this series coming to end, we can finally move on to preparing and transformation Agrobacteria tumefaciens on our journey to self-learn DIY Genetic Engineering of mushrooms and other fungi!
