Description and Learning Objectives: Repeatedly engaging in alcohol and drug intake despite personal, family and community harm is a hallmark of addiction. Modern neuroscience has used animal models of voluntary drug taking (i.e., self-administration) to define key brain circuitry that at first adapts to and then likely promotes addictive behaviors. One of these brain circuits appears to be integral to propagating habitual drug use, while another ties in emotional responses to perpetuation of addictive behaviors. Identifying the dysregulation of these pathways in addicted individuals is becoming possible with modern imaging techniques and targeting these pathways with a novel gene-targeted tools may uncover new approaches to preventing or treating drug addiction. Further, self-administration models also allow the development of valid and reliable biomarkers that that can help a person understand if their level of drug or alcohol consumption is harming their health or to monitor treatment compliance and/or effectiveness. Overall, new approaches to understanding the brain mechanisms of drug addiction is advancing preventative and treatment approaches to help improve our public health.
