Extracellular matrix, fibroblasts, multiphoton microscopy, collagen fibers
Video of Z-stack depicting primary human fibroblasts (red) and their self-assembled extracellular matrix (cyan) acquired using a multiphoton microscope. The fibroblasts were grown for 5 weeks on cell culture inserts with a pore size of 0.4 µm and the matrix secretion was stimulated by supplementing the medium with ascorbic acid. After fixation with paraformaldehyde, punch biopsies of the matrix were prepared and imaged. The fibroblasts were auto-fluorescent due to the ascorbic acid treatment, and the extracellular matrix was visualized label-free using second harmonics generation.
