Hi In case of MDR CoNS/methicillin resistant can we still depend on novobiocin to defrentiate bet. saproph. & epiderm.? In other wards for MRSE it would sensitive to novobiocin? Thanx

Hello. When you work in Lab hospital, THERE ARE SEVERAL TESTS THAT HOSPITAL MAKES FOR PATIETS. You have many but if you can show more, I just say -thank you. I have completed MT 3 years ago and finally found great gob. I am more competent in Chemistry and Hematology. For 3 years I have a little bit lost some information and now trying to remember . The theory is not big deal to remember, but with real tests- different .Watching you video I can refresh my memory fast1 Thank you

hi, im in a microbiology class and I am trying to figure out an unknown bacteria that i was given. it is a Gram + cocci and catalase +. the next test i have to do is the novobiocin test. my lab doesnt have novobiocin at the moment. does anyone know if i can use 10 micrograms of streptomycin instead of novobiocin?

Babak Sheikhpoor Hi Babak, I have never had to use anything else but novobiocin, so I don't know the answer to your question. What does your instructor say? Does your laboratory/class have procedures for identifying bacteria?

Patrick Tracy Hi Patrick, The assignment is to figure out an unknown bacteria. I have gone through all the steps on bergeys manual(gram stain, catalase test, mannitol fermentation, glucose fermentation) following what it is telling me to after each result except for novobiocin. My instructor says I need to find an alternative test because we don't have novobiocin. So I was just wondering, Because both novobiocin and streptomycin attach to the 30s ribosome, they would get the same result. Do you think I would have to do tests to seperate micrococcus bacterium from staphlococcus, like urease and nitrate? Also, thank you so much for even responding back to me. I can give you more information when I go back today and check my glucose and mannitol ferments today. Thank you again

Is the source of the bacteria human? If yes, from which body site? The body site usually tells you how far to go for identification. For example if we had a urine culture with a Gram-positive coccus that is catalase positive and coagulase negative, we would do the novobiocin to identify it as S. saprophyticus, but if the source is a wound culture, we may just stop after the coagulase test and call it a coagulase-negative Staphylococcus. If you don't have novobiocin, it is possible that you could rely on the mannitol-salt agar (MSA). Both S. aureus and S. saprophyticus grow on MSA and ferment mannitol (it will be yellow), but S. aureus will probably be beta-hemolytic on blood agar and will of course be coagulase positive, so it should be easily differentiated from S. saprophyticus. S. epidermidis will grow on MSA, but it will not ferment mannitol (it will be pink). Micrococcus spp., according to Bergey's, should have a yellow colony. Is the colony yellow like Micrococcus, golden like S. aureus or white like S. saprophyticus? I do not know how much testing you have to do to be able to identify your organism. This will depend on the source of the organism and your instructor. What I have written above is what I would tell my students, but you will have to rely on your instructor. Pat

Patrick Tracy The culture is from an unknown source. So i cant really answer that question.i tried the MSA test, and i found no change. it stayed a pinkish red, so maybe it did grow, but not ferment like you said.. I get more results from my urease test, blood coagulase test and my MR reaction results tomorrow. so i will use those to help me figure out what i have. i am going to ask my instructor tomorrow if there is anyway to get the novobiocin, i feel like i really need it to figure out my unknown. but anyways, thank you so much for your help. you were a god send.

Babak if you are following the Bergey's flowchart for catalase-positive, Gram-positive cocci, you don't have that many choices.

I thought novibicin test was done on nutrient a gar.The clear a gar.

I have never seen it performed on nutrient agar, so I can't answer your question.
