Hi Rimon, my understanding from David Sinclair is that resveratrol needs to be taken with fat for better absorption - he takes his with yogurt. Also trans-resveratrol is supposed to have better absorption. So the 200+ studies you have read and commented on, have you taken this into account, as this will significantly impact the results of the studies? My understanding is that most studies that say resveratrol has no benefits, are those that are not taken with fat.

This makes a lot of sense to me, and you know how picky I can be 😌👌 The only thing I would look into further is bioavailability and poor absorption of these compounds. Meaning, with low dosing it has to be so precise - and lipo forms do get absorbed so much better. So in that case should the dose be reduced even more? I would really like to know what Resveratrol are you taking yourself, what form/brand
@WellnessMessiah
2 жыл бұрын
Hey A T you're picky because you're smart. Yes, you're right about lipo. The problem is most studies just use regular resveratrol/fisetin/quercetin. If you see interesting studies on liposomal res/fis/que, maybe we can try to extrapolate the effect of the others 2, comparing lipo to non-liposomal. Their absorption is very similar. Resveratrol has the weakest solubility out of the three one less OH group) but for some reason, it's extremely potent nevertheless. Liposomal may even penetrate new tissues (e.g. hypothalamus, white blood cells). Feel free to provide me studies you see over times.

First of all, these are not boring, keep them coming! This was a very good video, and it confirms what I suspected, that resveratrol isn't a good senolytic. As I have said before, I take a total of 225mg daily, with 100mg being conventional resveratrol and 125mg being liposomal. This technically puts me in what you consider the medium range, but over half is liposomal, so presumably it reaches a lot more tissues and goes deeper into the cells. I also take quercetin every day, 250mg 3 times a day in phytosome form. Besides senescent cells there are benefits to quercetin like reduced inflammation. I know daily doses go as high as 1000mg, and I would guess that anything above that would put you in possible senolytic territory, which means I want to stay below that threshold. It also works as a zinc transporter which can boost your immune system if you're taking zinc (which I do). Recently I got some liposomal fisetin and began the Kirkland protocol, however I added liposomal 95% standardized OPC grape seed extract since pcc1 has shown evidence of being an excellent senolytic. I took these three days in a row, at 450mg fisetin and 600mg grape seed extract (I know the fisetin is below the Kirkland protocol, but my guess is liposomal delivery means you can go with less). However, after watching this video, I'm not sure how much benefit I can expect since I'm aged 45 and it looks like you need to be about 60 before you really need to kill off senescent cells. We definitely need more low dose fisetin studies. The liposomal pills I take are 150mg, so it would be nice to see what that dose would do. And we also need a LOT more studies looking at liposomal supplements. The few I've heard of show it makes them much more effective.

I'm not sure that Q and F are used togther so will recheck trial protocol. I alternate between F & Q (every 2 weeks) and divide into two doses per day. I am also not sure if doses are given as a bolus (all at once), or spread out over the day, and whether to use fat or black pepper to help with absorption. I take with a little fat (black olives or cocoa beans) I started donating plasma once a month (25% of plasma volume) at my local blood bank based Conboy studies, which among other things showed reduction of senescent cells in the brain. Very important topic, so hope ypu keep investigating and posting your findings. Thanks!

One thing that will boost absorption dramatically (maybe too much) is Soya Lecithin. One study showed that ingesting these polyphenols with soya lecithin increased blood concentration levels as follows: Quercetin X20, Fisetin X24, Resveratrol X29 There seems to be 2 factors at play here. The first is that lecithin is a phospholipid emulsifier and so helps in the absorption of molecules that are hydrophobic, as these polyphenols are. This is probably the main factor. The other is that lecithin can also protect the polyphenols so they pass through more of the gut, giving them more opportuntuies to be absorbed. It also helps to have a high concentration of Vit C in the blood - this seems to increase the time it takes the body to metabolize them. I mixed 500mg Quercetin powder, 300mg of Trans-Resveratrol and 500mg Vit C (in paste form - from a capsule) with 1000mg of Soya Lecithin, on a slice of wholemeal bread (also said to help absorption). After watching this video, I realize may have overdone it a bit with this combination (understatement). Thankfully, I appear to be fine and my kidneys are still functioning normally. I'm not taking anything at the moment but I think I'll adopt the hit and run approach in the future, and with much, much lower doses. Btw, soya lecithin also has a few of its own health benefits and is used extensively in animal feeds. That includes food for dogs, cats, fish, turkeys, chickens, cattle, pigs, etc. It increases nutrient absoption, stress resistance, regulates the immune system, significantly reduces bad cholesterol, reduces fatty liver deposits, boost bile production, among a few other things (can't remember the full list) - odd that we give to all our animals but not to ourselves. For obvious reasons, this combination requires a LOT more study.

I’m 60, a woman in great shape and a hectic life, that is, until 5 years ago when after a period of extreme workload collapsed and ended up with what is commonly known as ME. I’ve radically changed my lifestyle and doing “the right things”. Lately I’ve become very interested in the longevity aspect since longevity equals health. But I believe I may have reached an accumulation of senescent cell build-up due to that chronic stress I went through. My question is whether I should take Fisetin or Quercetin in a high dose for a short period of time and what would you consider to be a high dose (1g?) and for how long is a “short period”? I realise more studies are needed and coming up soon, but I would like to start before we have all the complete answers - at my own peril, as most of what I do is (conventional medicine has nothing for me). Would be so happy for any reply and this may be a question concerning a lot of people!

Resveratrol activates multiple sirtuins, we know oleic acid from olive oil is a far better sirt1 activator than resveratrol, but it is unknown whether it also activates the other sirtuins. Through splicing factors and also through sirt4, resveratrol lengthens telomeres and rejuvenates near senescent cells that are not damaged but have short telomeres. Resveratrol is also said to fight cancer at all three stages initiation, promotion and progression. It has lengthened the lifespan of all short lived species its been tried on. On longer lived species its effects on telomerase decrease with aging, younger animals are more rejuvenated. This could be due to NAD+ decline with aging in older animals. NAD+ boosters should in theory fix this and allow longer lived animals to also experience significant life extension, but this is a hypothesis.

A couple other things. You mention pterostilbene in this video. I think this would work best as a complement to resveratrol. From what I've read they activate different areas of SIRT1. So using both gives more benefits. Also, I seein future videos you mention olive oil and avocados. I'm guessing for oleic acid? This activates SIRT6 (as well as SIRT1) which needs more research! SIRT1 gets the fame for now but SIRT6 may be just as important or possibly moreso for longevity! Can't wait to see that episode! I've always tried to include olive oil in my cooking. Also love to eat avocados but man they go bad so quickly, it's much harder to incorporate them in to my diet.

Costco sell small Guacamole packages, that are individually packaged, and I’m guessing, have enough avocado 🥑 in each package, to satisfy a healthy daily requirement, whilst avoiding, having to toss out the over-ripe avocados…

Good job mate. But honestly have you seen Sinclair last year? He looks younger than before. He looks like 30 now. So I think all he does make sense, even though I agree with you low doses of resveretrol and quercetin and fisetina are not needed in our 30s. By the way have you found anything for combatting grey hair?
@WellnessMessiah

I agree he looks young. It could be his genes, his height (smaller people age slower, he's I think 5'6. Dwarf humans live commonly till 90 ), or the fact he has been cutting sugar in the last 20 years. I find it difficult to believe that high-dose resveratrol did it, although he could have genetic variation, which I hope I have too since I used to take his dose.

Soo question... I should switch to high resveratrol while fasting? Low otherwise. I aim for apoptosis while fasting, because elsewhere I read that the body is intelligent and during apoptosis targets plaques (e.g. Alzheimer's) and early cancel cells. FYI I regularly fast (only water+vitamins+minerals) for a couple of weeks every 3 months and lose 20 lbs (which I regain on purpose over the next 3 months).

what level of resveratrol in the blood does a dose of 120 mgs give you? if you look at the bioavailability of resveratrol and the plasma concentrations of free resveratrol form human studies you are not reaching levels of >25 uM which are needed to trigger the cell death effects. So 1 g of resveratrol should do this. You need to understand the PK of the drug before making a sweeping statement about the low dose that "in you opinion" give the best result. Not sure why you think you'd know more than Dr Sinclair on this, as he's extensively studied it and takes 1g.

Thank you so much, Rimon on this video. I did ask about Fisetin in light of dose dependency of Resveratrol. If I get you correctly, you think that 1.5g of fisetin in one day once per quarter for 70kg 55-60 year-old people? I know that nobody can tell until more solid studies emerge. I somehow feel that it’s too little. This strategy is acute stimulation instead of chronic which does make sense. However, why so different from Resveratrol of 100g every alternate day? 100g every other day also totalled up to 1.5g Resveratrol per month. Would like to understand why “chronic” intake of Resveratrol but acute intake for fisetin? And why not 1.5g in a day per month but per quarter? Could it because you’re being cautious in light of the Resveratrol surprise and that fisetin studies even more lacking? BTW, what do you think of spermidine?

I got sick from resveratrol making my blood pressure extremely high and I had to visist the ER several times before I understood that it was the resveratrol which made me sick. I took resveratrol every night with a high fat meal for a long peiod of time in doses between 600 - 1200mg and I've never been so sick in my life. My heart really worked hard and I got hypertension headaches & muscle weakness. I'm not a sensitive person at all and I've worked with supplements and taken alot for 10 years and never before got sick by supplements. I'm 31 and I'm a fully healthy individual who has never had high blood pressure before. I took resveratrol because nothing helped my tinnitus better than resveratrol for some reason. I'm not saying I believe it's necessarly bad just because I got side effects but definitly alot we dont know yet. Maybe Its better to take lower doses and less often as said in the video. This all happened 6 months ago and now when I took resveratrol 600mg just 5 days in a row again. I started to get some degree of the exact same symptoms back, which is scary, heavy chest, pounding heart, unregular heartbeats, diarrhea, weakness and terrible insomnia

There are other products as well using resveratrol from other sources, what I'd say is avoid low purity as most resveratrol comes from japanese knotweed which has significant emodin and emodin is bad for telomeres and emodin is also a laxative, high purity 98% or 99% is better.
