First person demonstration of how to subculture adherent BHK (Baby Hamster Kidney cells from ATCC) grown in T25 vented cap flasks. The spent media is removed; cells are washed with PBS, then 0.25 % (w/v) Trypsin-EDTA-PBS; incubated at 37'C for 2 min; cells are removed by slapping the side of the flask; fresh MEM is added to deactivate the Trypsin; 10 % of the original cell population is seeded back, fresh MEM is added to each; and cells are placed in the 5 % Carbon dioxide 37'C incubator using the proper technique to evenly disperse the cells along the bottom of the flask.
