Two questions came up for me: 1) What happens if we allowed the Strepp S gene from the HFR Bacterium to transfer to the F- Bacterium, can bacterial resistance get removed? 2) Does the genome from the HFR Bacterium always travel with the F+ factor first?

Only if enough genes move across to make it a complete prototroph. Or if you supplemented the minimal medium with the nutrients the cell couldn't make.

But in real time experiment, as we do not know what are all the genes (nutrient markers)present.....how can we choose the media for it Sir?

Trial and error with many plates and different nutrient combinations. The scientists that first did these experiments were very clever and diligent problem solvers. Today, we can sequence whole genomes and by using sequence homologies determine the order of genes. We've come a long way.

If you mean the new recombined strain, then yes it does grow. That is how we can tell which genes transferred.
