In this lecture, when you are explaining the technique of Isoelectric Focusing, you mention that a protein with a positive net charge would move towards a higher pH. This confuses me because I have understood, and correct me if I am wrong, that if the protein has a positive net charge then it has a "surplus" of H+ atoms, therefore, I would assume that the protein is acidic. I appreciate any explanation or clarification! Thanks a lot!

These are really good lectures. Just one minor tip, maybe try to move less. It's distracting from the lecture. The rest of it is amazing. Helped me really well for my test.

Sorry, one question. For Aspartate (acidic side chain). Why do we only average alpha-COOH and side chain? If the pH=3.6, then, alpha-COOH will be negative (pH>pKa) , side chain will be positive (pH

The side chain won’t be positive but neutral. COOH is neutral when acidic and negative when basic.

Why would IEF gel not denature protein? Since by going to their pI, you're messing with protein's charge (which in turn mess with their ionic & H-bond).

So theoretically if you placed a single unitary amino acid (the one you explained in step 1 where the pI would be 5.55) anywhere on the gel, the amino acid will migrate to its isoelectric point by the nature of the isoelectric focusing process - is this correct?

For case #2, best NOT to memorize but rather understand. Fully protonate at first (i.e., low pH) and then work your way up.
