If you are interested in how a radical is formed in Vitamin B12, it is formed by the homolysis of a Cobalt-carbon bond in Vitamin B12. The paper I looked at is by Dr. Jun Masuda, "How a protein generates a catalytic radical from coenzyme B12" in Cell Magazine.

very nicely explained. in the end when you discussed about vitamin B12 deficiency, the accumulated product methyl malonyl coA may result due to any of the two cases , either deficiency of vitamin B12 or mutation in mutase enzyme. then how is vitamin B12 deficiency particularly diagnosed?

You mentioned that a person who has a vitamin B12 deficiency would have an accumulation of D- Methanomalonyl coA, how would the latter product be produced by the propionyl coA if it were deficient of biotin? Thanks for the lectures :)!
