I found you because I was looking for the science I appreciate it actually I'm a medical doctor who graduated in 1995, so this information on the m TOR pathway was not available when I learned physiology, and since I don't work in primary care amd directly woth nutrition I really haven't learned about it . I have a personal interest in nutrition all my life as an athletic person and also wanting to stay healthy, but I'm trying to learn more about health , nutrition and review what I've learned on the past as I am now an Ayurvedic health coach.. so this is very helpful. thank you. your background is fine but between your little scenes when the screen jumping up-and-down ,,,that's annoying... you can change the transition and that will be more smooth. A blackboard or diagram even a simplified one will help, too? Otherwise, clear and concise.

Perhaps...but I'm not convinced that's necessarily a bad thing. I alternate between activating and not activating it. It's kinda hard to make any gains from working out without activating it. And I also take collagen peptides.

The key is periodic fasting for 5 days for authopagy and then eat all protein in 25 days.. according to leo and longevity if you fast for 5 days your IGF-1 will slow down for 15 days...

This video is very hard to listen to. Your voice is not very clear maybe you should get your voice box checked out for vocal polyps and why the background music it’s not needed and makes it worse. The background set is pretty depressing as well.

How can Perfect Amino claim zero calories? Amino Acids have Carbon. Methionine, for example, leaves the Methionine Cycle, via the Transsulfuration Pathway, and contributes to the TCA Cycle.

Whether or not is has calories depends on if it's used as energy (catabolic, then it does have calories) or whether it's used to synthesize new proteins (anabolic, no calories).

@@LanceHitchings A calorie is defined as the energy needed to raise the temperature of 1 gram of water through 1 °C. This measurement is determined in the lab not the body. What the body does with a calorie is another story and should be referred to as an "Effective Calorie".

Why not discuss how fasting fits in? Presumably, autophagy cranks up because there then is a lack nutrients. Thusly, mTOR would not be activated. Then, when I'm not fasting and doing HIIT, mTOR would be inhibited boosting AMPK allowing autophagy too. Could you comment here s'il vous plait?

Excellent vid as always. Thumbs up on the new set, you have a great eye. The obvious tools to mention are metformin and berberine. Not a fit for your program of cycling, but I believe many people feel they don't pose obstacles to your fitness goals. There is even evidence they work better together.

I'm most intrigued with the Mtor balance puzzle you present, but first, production comments: Nice basement studio, Lance. Nice new format, the CC is better positioned on my screen. Excellent use of infographics. Fun puzzle of what's the best mix of HIIT, weight training, amino acid supplementation and, in my case, my weekly dosing of Rapamycin? I'll be researching weight training recovery times, maybe doing the most resistance training 3 days after (and 4 days before) my dose of Rapamycin. Since I've been on Rapamycin (5 months) and increased my HIIT training, my resting heart rate is lower than it has ever been in my life - including my extremely athletic state at age 30. My athletic performance now is pretty good and I am tracking it. Please continue the science detail in at least some of your videos, perhaps researchable links, as well.

I enjoyed your video,and I think you might be on to something,with balancing the Mtor pathway in the manner you are suggesting,I dont do heavy weight training,I do however fast on average 16 to 18 hours a day,and do a mild form of HIIT training in the morning fasted.I have noticed at 62 years of age,some sarcopenia,but I am not too concerned with it,I am more concerned about extending lifespan,,I am following some of Dr David Sinclair's protocol,by taking Pterostilbene,MNM's Resveritrol MK7 and a few other things, I also am coupling this strategy with a form of relaxation and de stressing techniques to keep cortisol levels in check...thank you Ron Stanley.

Nice talk but you completely missed the boat on autophagy and ampk activation, which really only occurs during severe calorie restriction or, ideally, fasting >24 hrs. If you continue to consume aminos then you will inhibit autophagy.

For me, the jury is still out on this...but I'm leaning in your direction. I've heard that a 72 hour fast is required. But I've also heard of techniques for staying in a state of mild autophagy on a somewhat permanent basis. And that there's a limit to how many aminos you can consume and still stay in some state of autophagy, but it's not none. It's such a new concept, I think there's still a lot to learn.

Trehalose can produce autophagy independently of mTOR, so autophagy can be produced without mTOR inhibition. But I’m in psych and I was curious about something. Increasing MTOR signaling stimulates neuritogenesis, this is how psychedelics grow ur brain, how do we activate mTOR neuritogenesis while increasing longevity. Can we inhibit mTOR for most of the day but then stimulate it when we are in that really hard class? What are the durations of mTOR activation?

I started at 330lbs i am at 265ish now in 5 months.. now I'm starting to build alot of muscle ... I got 11B mTOR Active to help my gains.. now after watching this im not sure lol .. I am still at a calorie decrease of 1200 or less daily.. I go to gym everyday for 1 to 2 hours... should I not take these ?

I think the key is to balance mTOR with AMPK. Both are beneficial but since you can have one or the other, but not both, you need to cycle between them. I don't think mTOR is that bad if you balance it with a period where you fast, undergo autophagy and activate AMPK. I'm working on a series of videos about this very subject and I'll be addressing balancing mTOR and AMPK in a future video. But yeah, if you do nothing but activate mTOR all the time by always trying to build muscle, that could work against you. Just my opinion, I'm not a doctor.

Kinase is KY naze What you propose is consistent with what David Sinclair of Harvard, a major anti-ageing researcher (I’m sure you’ve heard of him) and others seem to be saying. I think this is a very complex subject and while I am following a similar regime I think the current picture will develop with time. Current thinking may then appear slightly naive. But that’s inevitable of course - humans can’t be researched like lab rats/mice. Until then I would like see the importance of the micro-organisms in our diet/biome and diurnal rhythms of optimal fasting integrated into the scheme by experts.

Here's my take: If we could live to 200 years, say, then the problems with the planet that we're leaving to our children and grandchildren and their children suddenly become OUR problems. I think that would wake a lot of people up. Also, among 1st world countries, many of them have a negative birth rate. Their populations are getting smaller, not bigger. That's just a couple of thoughts, there's more. I think longevity could be the answer we're looking for.

You do know that I'm 71, right? And for me, it's not how good I look, it's all about how young I feel. For whatever it's worth, I've taken about 10 different biological age tests, and they put my average biological age at around 55. So, I kinda feel that whatever I'm doing, it's working great!

Also also Also. Mtor Vs AMPK aka cell growth vs cell apoptosis or atophogy. But how do u feel about BUILDING muscle while also getting the benefits of AMPK? Both can’t be done at the same time. I try to activate MTOR in the morning by Aminos and workouts but in the afternoon I restrict calories or Fast for the AMPK but im having a hard time trying to find the sweet spot when it comes to timing for both.

To be honest, I currently have no idea of how to find the sweet spot. I'm currently cycling back and forth between the two, but doing it every day or two, rather that between morning and afternoon. But I have no way of knowing how that's working. If you have any info, please share.

berberine only inhibits the mtor partially, not completely. correct? will I still get good benefit from Hiit while taking berberine?

Although I'm unaware that berberine can diminish the effects of mTOR, it's possible that it may very well. I've had people comment that berberine can impede cardio performance, although I haven't experienced that myself.

(Basically retired now, his daughter and her husband have Peace Seedlings) He has done a lot of research on for plants and plant communities, including the abundance of "other" tubers from Peru (beyond potatos) ....Alan comments that the pods of both green peas and beans have well absorbed amino acids -

Great discussion but I don't think this will work man. Your muscle growth and repair doesn't just take place on the days you weight train, certainly not only 2 hours after when you take your amino drink. You do your most growth and repair on the days you rest. Also, while HIIT training on your non weight training days activates AMPK instead of MTor, you are still damaging muscle tissue and you need protein for repair as well.

i have 24 h a day i fast 16 h and eat for 8 hours , i know all mtor inhibitors and use them daily but how can i balance my lifestyle so the body will rebuild and repair tissue with what i eat and drink and when to shut down this process in order to build muscule and live properly , i am making mistakes with IF i know that because i keep losing muscle mass get ROS attacks lose mitochondria

That's a great question, and one I struggle with myself. I too want to build new muscle tissue, which require activating the mTOR pathway. What I do is cycle between strategies that activate the mTOR pathway and the AMPK pathway...but I don't know how to test to find out if it's actually doing any good. Good luck, and let me know if you find a definitive answer to your question. We're all in this together.

Very good physiological information! In one of Dr. David Sinclair's talks he mentions Metformin and Green Coffee Extract both activate the AMPK pathway. Presumably alternating between resistance training/essential amino acid days (anabolic days) and Metformin or Green Coffee Extract and less amino acid days (catabolic/autophagy) with rest would be one method of pulsed therapy. Dr. Sinclair emphasizes that no one knows yet what the optimal regimen is for this kind of anti-aging therapy. It would be great if people pursuing it would keep records and do either the DNA methylation blood testing or telemerase length testing and contribute their data to some centralized database so that we could determine what the optimal protocol is. I believe William Faloon's (of The Life Extension Foundation) non profit organization is compiling such a database but I forget the name of his non-profit at the moment.

I agree, it's really difficult to know what the most efficient protocol might be. I'm sure that THEY (the researchers and scientists) will get it figured out one day, but in the meantime, it's up to us bio-hackers to experiment and figure it out on our own...by stay current on the latest literature and keeping great records. Last weekend (Oct 3-6) I attended RAADFest, in Las Vegas. It was put on my the Life Extension Foundation, and the Coalition For Radical Life Extension. Bill Faloon announced a new study into age reversal, and signed up participants. I signed up, and I believe that they're still enrolling new subjects, although I can't find any info yet on how one could enroll. So, I think this is one way the Life Extension Foundation is compiling a database on aging.

Smart guy. All reference data he uses is consistent with what I've learned from multiple other credible sources. His conclusions about interpretation of the data and actions to take make sense.

Based on the science of mTOR and AMPK, what would you think of an alternating stack for fasting- NAD+ during the fast and Berberine or even better, Dihydroberberine during the feeding window?

That's a really interesting proposal. To be honest, I don't really have an answer. I guess I'd like to hear your reasoning behind that suggestion before I offer an opinion.

@@LanceHitchings I'm thinking NAD+ would support mTOR pathway progression. It could also increase sirloin levels. Berberine would support lower blood sugar levels, increases insulin sensitity and opens the AMPK pathway, increasing fat metabolization

Hi Lance, really informative video. Personally really like your balance of science and recs. Question for you - given that rapamycin "tricks" the body into thinking that there's less caloric availability and inhibit mtor, what would happen to calories if you took rapamycin while having caloric abundance? It sounds like autophagy recycles existing amino acids so I'm curious what would happen to new calories in this case. Thanks!

Shutting down mTOR is something that happens in response to fewer calories. Rapamycin causes a similar response, but it doesn't effect how those calories are handled. If you take rapamycin while having a caloric abundance, you're going to gain weight, just like you would if not taking rapamycin. I think. I'm not a scientist or a doctor, so that's just my opinion.

On the late freight here, but will give my views anyway. Yes on the new studio. I like it. Also an absolute yes on the science. Like many other commentators I am impressed with your ability to make it understandable. Because the longevity and age reversal issues are so complex I fewl it is very helpful to build up an understanding of the science that supports the superstructure of the activities such as IF, HIIT, and resistance training plus diet and supplements. Oh yeah, the science is important so please continue. Now for this video's content. I'm watching this in December, so a few months after posting. Are you still doing the on and off high low protein coupled with resistance training? In the video you mentioned the protein you take. I'll have to go back to get the name. Would you consider doing a video on why you chose this protein supplement.

Yes, I'm still doing the same routine with regard to protein. The protein I'm taking is called "Perfect Amino", and I'm doing a video on it in the next batch of videos that I do...so it should be out in about a month or two.

@@LanceHitchings I bought a tub of Perfect Amino and am taking it first thing in the morning. The sight said it wouldn't break a fast because it goes directly into the bloodstream. They didn't support the claim with any data. I'm buying a blood sugar test kit to check my blood to see what actually takes me out of the fasting state. I'll let you know what happens with Perfect Amino.

I know that phosphatidic acid is supposed to help with strength training, but since it has no apparent effect on longevity, I haven't really studied it. I know that it is supposed to activate the mTOR pathway...so I guess it might have a negative effect on longevity.

Hi, Lance. This is Max from Debbie's class at OCHS. I think you pretty well nailed the subject here. I don't really lift but I hit the Bowflex a couple times a week and I like to follow that with a big mug of hot chicken sipping bone broth into which I have added 20 gm of collagen peptides from cow hide. It has all the AA's except for tryptophan and best of all only 106 mg of Methionine. At 90 kg body weight my RDA for Methionine would be about 900 mg and this leaves me room for the egg, goat cheese, and fish in my diet. This seems to work pretty well for me. Liked the video. Now can you please explain how to cure cancer and balance the federal budget? Thanks.

I don't think the body stores a lot of energy in ATP. Yes the mitochondria make it: AMP --> ADP --> ATP but we don't have enough to store any amount of energy there. You stores of energy glucose, glycogen, adipose, and muscle protein in that order. I think of ATP/ADP/AMP more like a tennis ball getting whacked back and forth across the net. Pretty sure it turns over every minute or two. Otherwise you could hold your breath a LOT longer.

@@LanceHitchings Lance, I like what you're doing here. A little personal history - after the Army I went back to college and got my degree in Biochemistry in 1979. (Thanks, GI Bill.) Wound up spending most of my career in IT designing software so I didn't really keep up with all the research going on during the last 40 years. Now that I am retired and 'aging' I am fascinated, but not surprised, by all the discoveries that have been made. The more I learn about the various biochemical pathways that have been elucidated and the behavioral strategies designed to exploit them the more I am convinced that it's all about diet, exercise, stress, sleep, and relaxation and finding the proper balance. Beyond that it's mainly our individual genetic defects and the hazards of living in a less than pristine environment. Big pharma has produced some very useful drugs over the last few decades but increasingly those guys are all about producing pills that earn them billions by enabling us to keep from reforming our bad lifestyle choices. I'm sorry to tell you that David was killed in an automobile accident back in 1990 along with Mark and a friend of theirs. Mark raced in the Daytona 200 that day with David serving as his mechanic. They were on there way to their motel just north of the city and never made it. I can't tell you how much it hurt to get that phone call from my mom telling me that my brothers were gone. Watching my parents over the next few weeks also taught me a lot about how damaging real stress can be to you both mentally and physically. They both aged about 10 years in a few days. My dad suddenly had to use a cane to walk and my mom went gray almost over night. I think losing a child must be the worst thing that can happen to you. Losing two....

About your PerfectAmino: Looks like a good product but don't kid yourself about it having Zero calories. Eating amino acids is like eating protein except that your body doesn't have to expend energy to separate them from the protein chain. You would actually get more calories that from eating protein. Your body does require those essential aminos exogenously to build muscle but it can also metabolize them as fuel so they do have calories. Even if you could incorporate 100% of them into muscle mass they will still turn over with time. Kinda like capturing CO2 in plants. After a while it goes back into the atmosphere.
